Growth Inhibitory Effects of the Ethyl Acetate Soluble Fraction of Perilla Frutescens by MTT Assay in Human Oral Epithelioid Carcinoma Cell and Human Skin Malignant Melanoma Cell

Abstract

|
The cytotoxicity of the ethyl acetate soluble fraction of Perilla frutescens on human oral epithelioid carcinoma cell and human skin melanoma cell lines was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-Biphenyl-2H-tetrazalium bromide (MTT) colorimetric method. The light microscopic study was carried out to observe morphological -changes of cultured - human oral epithelioid carcinoma cell lines (KB) and human sldn melanoma cell lines. (SK-MEL-3 Cells).
These results were obtained as follows ;
1. In MTT assay, subfraction 1 and 2 were shown significant cytotoxicity at 10mgtml concentration.
2. The comparison of IC5o values of various fractions in human oral epithelioid carcinoma cell and human skin melanoma cell lines was exhibited that their susceptibility to theses fractions decreased in the following order: human oral epithelioid carcinoma cell lines: subfraction 5 < subfraction 4 < subfraction 3 < subfraction 2 < subfraction 1, human skin melanoma cell lines: subfraction 5 < subfraction 4 < subfraction 3 < subfraction 1 < subfraction 2 by MTT assay. In light microscopy, the subfraction 1 and 2 of the ethyl acetate soluble fracion of Perilla frutescens showed the highest cytotoxicity. These findings suggest that fracion 1 and 2 possess the most antitumorous agent.
